The earliest I have info of Isaac is a Tithables list dated 1741. Does anyone have any earlier record of Isaac or know of his parents? Would love to share info.

I think the Isaac that you are researching is very likely Isaac Duffer of Amelia, Cumberland Cty VA. He was born abt 1733 and married Mary Vaughn. You can find him in the IGI files and the Ancesteral files at: http://www.familysearch.org/
I am researching him as well and so far I have not been able to confirm any parents for him. I have found a John Duffer of henrico cty VA brn abt. 1695 listed in the pedigree resource files at the above site. It seems likely that they are related.
I hope this was helpfull, please let me know if you are able to find anything more on Isaac.

We are researching the same Isaac. I know of no one with primary source evidence of the Vaughn marriage. Isaac's wifes name was in fact Mary (from his will and land records). The assumption she was a Vaughn comes from the will of Henry Vaughn who mentions the children of Isaac Duffoe as heirs. I have been unable to find any marriage record (bond or minister's return). I also have seen the John Duffer of Henrico Co. and his will mentions his wife and no children or grandchildren. The earliest record I have of Isaac is a tithable list of George Booker in Amelia Co.

There is a will for a John Duffee/Duffy in Amelia Co., which I have yet to obtain a copy or abstract of (is on my todo list). According to the index to Amelia Co. Wills and Administrations (1734-1800) the will was probated 21 May 1742, accounts dated 30 Nov 1742, and inventory and appraisal recorded 16 Jul 1742. It is in Will Book No. 1, 1734-1742, according to the Library of Virginia.
