My 6th great-grandmother was Lavina Brandenburg (b. 1809, Kentucky, USA), grand-daughter of Matthias Brandenburg (b. 1740, Brandenburg Germany), whose 2nd great-grandfather was George W Hohnezollern (b. 1595, Prussia). I'm not sure if this is what you are looking for, but Lavina Brandenburg's grandchild, James B Grable (b 1856, Missouri, USA) married Lavina Kinder (b. 1858, Missouri, USA), who were rumored to be 1st cousins. They had six children; two of those children had cleft palates and one of their children without a cleft went on to have a child with this recessive genetic disorder. Let me know if this is the gene to which you are referring.
