James Stuart, Earl of Moray was the illegitimate son of King James V of Scotland and Margaret Erskine, dau. of John, 4th Lord of Erskine.
He was sent to France to invite his half sister, Mary, to return to Scotland and rule. On her return he acted as prime minister and advisor. In 1562 she created him Earl of Mar, but that title being claimed by the Lord Erskine, she created him instead Earl of Moray in 1564.

He strongly opposed Mary's marrriage to Lord Darnley (1565) and openly appealed for a call to arms, and soon afterwards had to flee to England. He did not return to Scotland until after the murder of Rizzio, in which he was probably privy. In Arpil 1567 he withdrew to France but was recalled by Scotish nobles in August to take up arms against Mary. Upon his return he found Mary a prisoner at Lochleven and appointed himself Regent. Upon Mary's escape, he defeated her forces at Langside, near Glasgow (May 13, 1568) and on Jan. 20, 1570 he was assasinated at Linlithgow.

Mary was daughter of James V of Scotland and Mary of Guise, Princess of France. James V was son of James IV of Scotland and Margaret Tudor, sister of Henry VIII of England.

Mary of Scotland was married three times:
1st in 1558 to Francis, dauphine of France (later King Francis II) who was her 5th cousin and who died 1560.

2nd m. 1565 to her half 1st cousin, Henry Stuart, Lord Darnley who died 1567. He was son of Matthew Stuart 13th Earl of Lennox and Margreat Douglas who was daughter of Archibald Douglas and Margaret Tudor (widow of James IV of Scotland and mother of James V)

3rd m. James Hepburn, 4th Earl of Bothwell

By her 2nd marriage to Henry Stuart she had a son, James Stuart who became James VI of Scotland and later James I of England.

James I of England did receive John Rolfe and his wife, Pocahontas, dau. of the chief of the Powhaten people.

I do not think there was any family relation between King James Stuart and the Rolfe family.
